FOXO4-DRI is a synthetic, cell penetrating peptide derived from the FOXO4 protein and designed to interfere with the interaction between FOXO4 and the tumour suppressor protein p53. This pathway is involved in the survival of senescent cells, which are cells that have stopped dividing but remain metabolically active. FOXO4-DRI is being investigated as a senolytic peptide for its potential to selectively promote the removal of senescent cells, making it an area of interest in cellular ageing and longevity research.

Research

Research has demonstrated that FOXO4-DRI can disrupt the FOXO4-p53 interaction and promote apoptosis of senescent cells in laboratory and animal models. Studies in aged mice have reported reductions in senescent cell burden alongside improvements in measures of physical function, tissue homeostasis and age associated changes. Further research has investigated its effects on senescent endothelial cells, chondrocytes and other cell types, with recent structural studies providing additional insight into how FOXO4-DRI interacts with p53.
